How to Prepare Alkaline Lysis Solution II (P2)

Preparation Strategy

Method: stock_then_dilute

Reason: NaOH mass (8g) exceeds 1g threshold, but stock approach recommended for accurate alkaline solution preparation and safer handling

Procedure

  1. 1Put on safety equipment: nitrile gloves, safety glasses, lab coat, work in fume hood
  2. 2Add approximately 800 mL of distilled water to a 1 L beaker or flask
  3. 3Slowly add 8.00 g NaOH while stirring continuously - solution will become very hot
  4. 4Allow solution to cool to room temperature (15-20 minutes) while stirring occasionally
  5. 5Once cooled, slowly sprinkle 10.0 g SDS powder onto the surface while stirring gently to minimize foaming
  6. 6Continue stirring until all SDS is completely dissolved and foam subsides (10-15 minutes)
  7. 7Transfer to 1 L volumetric flask and adjust to final volume with distilled water
  8. 8Mix thoroughly by inversion
  9. 9Solution should be clear and colorless - any cloudiness indicates incomplete dissolution

Sterilization

Filter through 0.22 μm filter if sterile solution required. DO NOT autoclave - SDS will precipitate at high temperature and NaOH concentration may change due to CO2 absorption.

Safety

EXTREME CAUTION: Highly caustic solution. Wear nitrile gloves, safety glasses, and lab coat. Work in fume hood. NaOH generates significant heat when dissolved - allow cooling before adding SDS. Can cause severe burns to skin and eyes. SDS powder can cause respiratory irritation.

Storage

Store in tightly sealed plastic bottle at room temperature. Label with preparation date. Solution stable for 6-12 months. Do not store in glass long-term as NaOH etches glass. Keep away from CO2 exposure which will reduce alkalinity over time.
