How to Prepare 10x PBS Stock Solution

10X concentrated PBS stock solution, dilute 1:10 for working solution

Procedure

  1. 1Add approximately 80 mL of distilled water to a 100 mL volumetric flask or beaker
  2. 2Add 8.01 g NaCl and stir until completely dissolved
  3. 3Add 0.20 g KCl slowly while stirring to prevent localized supersaturation
  4. 4Add 1.42 g Na2HPO4 and stir until dissolved
  5. 5Add 0.24 g KH2PO4 and stir until dissolved
  6. 6Adjust volume to exactly 100 mL with distilled water
  7. 7Mix thoroughly by inversion or stirring
  8. 8Verify solution is crystal clear before use

Sterilization

Autoclave at 121°C, 15 psi for 15 minutes. Allow to cool before use. Solution is stable during autoclaving.

Safety

Level 1 safety: Standard laboratory gloves, lab coat, and safety glasses. All components are non-hazardous salts.

Storage

Store at room temperature in tightly sealed container. Stable for 1 year. If crystallization occurs during cold storage, warm to room temperature and redissolve before use.
